The problem you may encounter would be the maximum folder indexing. Most head units can only read/index up to a certain number of folders or files.
My old JVC unit wouldn't read any more than 99 folders (including subfolders etc).
No harm in trying - but I would delete anything other than music files on the hard drive before trying.

DNY*BOY: I take it that you're using external HDD that is powered through the USB adapter?
I've got 2x external hard drives but they both have 240v power plugs (to power them) as well as USB cables (for connectivity).
Is the ext HDD powered through the USB port? Wouldn't that pull alot of power through that port!? :|

generally the 3.5" externals are 240v powered but the 2.5" drives are usb powered - however the port on the HU may not be powered and in any case, some drives (eg my 80gb maxtor) require TWO powered usb ports to run (stupid i know)
it does work, but as someone correctly pointed out, in some cases there is a limit to the number of folders that can be read
